Reaction of Different Inoculation Densities of Meloidogyne incognita race 2 on Some Resistant Pepper lines

Abstract

Reaction of
different inoculation densities of two Meloidogyne
incognita
race2 isolates (G7 and K4) was
studied on the root-knot nematode resistant pepper lines Carolina Wonder, PM217,
CM334 and susceptible Yolo Wonder
under controlled conditions (25±2°C temperature, 60±5% humidity and 16:8
photoperiodizm). Experiments were conducted on a randomized block design with 5
replications. Egg masses were used inoculum source and when the plants reached
4 real leaf seedlings, 1-5-10-20 and 50 egg masses were inoculated into the 2
cm depth of plant root zone. Peppers were harvested 9 weeks after inoculation
and root gall ratio was indexed according to 0-5 scalae. Numbers of galls, egg
masses and eggs in a roots were counted under light microscopy and reproduction
fitness were evaluated. G7 and K4
isolates showed avirulent reaction in all inoculum densities in Carolina Wonder
and CM334 resistant pepper lines. In the PM217 resistant pepper line, Reproduction
fitness of G7 isolate was found 1.1 in the density of 50 egg masses inoculum
densities, and the durability of the pepper line is broken.10-20 and 50 egg
mases inoculum densities of K4 isolate showed virulent reaction in the PM217
resistant pepper line. Reproduction fitness in these inoculum densities were
1.4, 3.5 and 4.4 respectively. The
pathogenicity of both isolates in the Yolo Wonder pepper line was found to be
high.

Meloidogyne incognita ırk 2'nin Farklı İnokulasyon Yoğunluklarının Bazı Dayanıklı Biber Hatlarında Reaksiyonu

Abstract

Çalışmada, Meloidogyne incognita ırk 2'nin iki izolatı (G7 ve K4) 'nın farklı inokulasyon
yoğunluklarının
Kök-ur nematodlarına dayanıklı Carolina Wonder, PM 217,
CM334 biber hatları ile M. incognita
'ya duyarlı Yolo Wonder hattında reaksiyonu kontrollü koşullar altında (
25±2°C sıcaklık, %
60±5 nem ve 16:8 fotoperiyot)
araştırılmıştır. Denemeler, tesadüf blokları deneme desenine
göre 5 tekerrürlü olarak kurulmuştur.
İnokulum kaynağı olarak yumurta paketi kullanılmış ve bitkiler 4 gerçek
yapraklı fide dönemlerine geldiklerinde
bitki kök bölgesinde açılan 2 cm derinliğe 1-5-10-20 ve 50 yumurta paketi inokule edilmiştir.
Biber bitkileri inokulasyondan
9 hafta sonra sökülmüş ve köklerdeki gal oranı 0-5
skalasına göre indekslenmiştir. Köklerde gal sayısı, yumurta paketi ve yumurta
sayısı ışık mikroskobu altında sayılmış ve üreme gücü hesaplanmıştır.
Carolina Wonder ve CM334  dayanıklı biber hatlarında G7 ve K4 izolatı
tüm inokulum yoğunluklarında avirülent reaksiyon göstermiştir. PM217 dayanıklı
biber hattında G7 izolatının 50 yumurta paketi inokulum yoğunluğunda üreme
oranı 1.1 olarak bulunmuş ve dayanıklı biber hattında dayanıklılığı kırdığı saptanmıştır.
K4 izolatı PM217 dayanıklı biber hattında 10-20 ve 50 yumurta paketi inokulum
yoğunluğunda virülent reaksiyon göstermiştir. Bu inokulum yoğunluklarında üreme
oranı sırasıyla 1.4,3.5 ve 4.4 olarak bulunmuştur. Yolo Wonder biber hattında
her iki izolatın patojenitesi yüksek bulunmuştur.
